It is in direct correlation with the prevalence of the metabolic syndrome and the western lifestyle. Thus, we aimed to study the impact of fatty liver disease globally, depending on geographic distribution, age, gender, and associated risk factors, using the searche engines PubMed and Google Scholar. It has been observed that steatotic liver disease is a globally prevalent disease, which is more common in men and post-menopausal women, suggesting the protective role of female sex hormones. Steatotic liver disease is also considered the hepatic manifestation of the metabolic syndrome, being frequently diagnosed among patients with type 2 diabetes and\/or obesity. The increased incidence of the disease was also noted in children, overweight \/ obese adolescents. In order to address research tasks, a combination of theoretical and empirical methods was employed: analysis of psychological and pedagogical literature, comparison, systematization, specification, generalization of pedagogical experience, questionnaire surveys, testing, and pedagogical experimentation. The diagnostic tools utilized were: the Achievement Motivation Questionnaire (adapted by M.Sh. Magomed-Eminov) and the Assertiveness Questionnaire (V.P. Sheynov).\r\nResearch results and conclusions. The definitions of such scientific concepts as \"assertive behavior,\" \"assertiveness,\" \"assertive personality,\" and \"achievement motivation\" have been specified; the components of assertive behavior have been examined. At the stage of summarizing the results of the conducted research, students were grouped according to the identified achievement motivation, enabling a comparison with the level of assertiveness characteristic of each respondent. The obtained data have been statistically confirmed. The project entails three goals. The first goal involves establishing a fluorescence-based system to detect individual interactions between aggregates and proteasomes. To achieve this, I will initially receive training on proteasome purification and examine how to modify proteins with specific ubiquitin chains. Subsequently, I will explore different fluorescence labe lling systems and optimise a labelling strategy for protein aggregates and the proteasome without disrupting their integrity. The fluorophores selected should be bright and photostable to be compatible with the single-molecule instruments. This single-molecule fluorescence system will be used to establish a method that can reveal interactions between individual protein aggregates and a single proteasome to determine the efficiency of degradation for different types of aggregates. The second goal will focus on aggregate degradation in cells using three-dimensional superresolution imaging. Heterogeneous interaction between aggregates and proteasomes can be resolved to determine whether certain types of aggregates can inhibit the proteasome. Finally, I will focus on cell lines with mutations on key Parkinson's disease-related genes, and establish whether aggregate degradation is affected. I will use a powerful experimental strategy that combines cellular-resolution calcium imaging, behavioural analyses, optogenetic circuit manipulations, neuroanatomical tracing and computational modelling to discover how brain-wide circuits operate at the cellular level to flexibly control the expression and coordination of behaviour. We report a novel orthorhombic crystal form of DNA-free\n Thermoplasma acidophilum<\/jats:italic>\n XPD (TaXPD) obtained under high ionic strength conditions generated by sodium potassium tartrate and NaCl-based vapor-diffusion conditions, in contrast to earlier previously reported conditions that used polyols (PEG) or diols (MPD). The crystals belonged to space group\n P<\/jats:italic>\n 2\n 1<\/jats:sub>\n 2\n 1<\/jats:sub>\n 2\n 1<\/jats:sub>\n (\n a<\/jats:italic>\n = 59.53,\n b<\/jats:italic>\n = 96.00,\n c<\/jats:italic>\n = 159.09\u2005\u00c5) and diffracted to 2.13\u2005\u00c5 resolution, yielding the highest resolution TaXPD structure to date. Structural analysis showed that this crystal form contains fewer intermolecular interfaces than the previously reported hexagonal lattice, as supported by\n Protein Interfaces, Surfaces and Assemblies<\/jats:italic>\n (\n PISA<\/jats:italic>\n ) analysis. This supports the determination of the 510\u2013514 loop in the long-form DNA-free TaXPD, which was previously disordered in other structures. This work highlights how crystallization conditions influence lattice organization, structural completeness and diffraction quality.
