Populacija mu&amp;scaron;karaca koji imaju seksualne odnose sa mu&amp;scaron;karcima(MSM) predstavlja posebno osjetljivu grupu za prenos HIV-a i drugih polnoprenosivih infekcija.Vulnerabilnost se pove\u0107ava usljed rizi\u010dnog pona&amp;scaron;anja,marginalizovanja, stigmatizovanja kao i nedostupnosti zdravstveno-socijalneza&amp;scaron;tite. Cilj rada je bio da se procijeni prevalencija HIV-a i odabranih polnoprenosivih infekcija te ispitaju znanje, stavovi i pona&amp;scaron;anje u odnosu na HIV\/polno prenosive infekcije u MSM populaciji u Bosni i Hercegovini, kao i dase dobijeni rezultati uporede sa istra\u017eivanjima iz 2008. i 2010. godine.Metode. Istra\u017eivanje je provedeno kao bio-bihejvioralna studija prevalencije uMSM populaciji. Obuhva\u0107ena su ukupno 333 ispitanika, u &amp;scaron;est gradova Bosnei Hercegovine. Istra\u017eivanje je sprovedeno u periodu septembar &amp;ndash; decembar2012. godine, kroz dobrovoljno, anonimno i povjerljivo anketiranje. Nakoninformisanog pristanka i obavljenog savjetovanja, uzet je biolo&amp;scaron;ki materijalza laboratorijsko testiranje na HIV, hepatitis B, hepatitis C i sifilis.Rezultati. Istra\u017eivanje je pokazalo prisutnost rizi\u010dnog pona&amp;scaron;anja u MSM populaciji:multipli partneri (prosje\u010dno tri razli\u010dita slu\u010dajna partnera u proteklihmjesec dana), neza&amp;scaron;ti\u0107eni seksualni odnosi (samo 20% ispitanika redovnokoristi kondom sa stalnim partnerom, 37% sa slu\u010dajnim partnerom, 4,6% prioralnom odnosu), seksualni odnosi pod uticajem alkohola (69,6% ispitanika)i droga (26,7% ispitanika) u poslednjih &amp;scaron;est mjeseci, seksualni odnosi i sa\u017eenskim osobama (45,5% ). Uprkos prisutnosti rizi\u010dnog pona&amp;scaron;anja, 14,8%ispitanika smatra da rizik od preno&amp;scaron;enja HIV-a ne postoji, a 41,3% da je nizak.Serolo&amp;scaron;ka ispitivanja pokazuju i dalje nizak nivo HIV\/polno prenosivih infekcijau MSM populaciji u BiH.Zaklju\u010dak. Iako postoji porast za&amp;scaron;titnog seksualnog pona&amp;scaron;anja i u\u010destalije HIVtestiranje u odnosu na ranije periode, to nije dovoljno za odr\u017eavanje niskognivoa infekcije HIV\/polno prenosivih infekcija. Therefore, the methodical approach in the interpretive processing of this complex and layered artistic literature material is a serious teaching challenge. The hypothetical starting point in this case is the phenomenon of dystopia present in the chosen reading books which is seen through literary and artistic processing. Through this topic, we have meticulously and seriously analyzed the relationship between the novel and the reading books, as a first and obligatory textbooks for teaching literature. The aim of our research is to determine the presence of novelistic dystopian art material in gymnasium's and secondary vocational schools' manners of interpretation. The subject of the research are secondary schools' textbooks in the Republic of Serbia present in teaching practice in the period from the first half of the XX century and in the first decades of the XXI century.We based the research on forty copies of reading books from all secondary education grades. The reading samples are works of thirty authors and are printed in several different-unmodified, modified, supplementary and revised editions - focusing on the most characteristic copies of reading books from different periods and system and program phases that significantly affected their content. We believe that the chosen sample of the study is reliable, and the conclusions we came to are a realistic presentation of the state of novelistic prose in our reading books. Although the novelistic text is only one literary reading segment, our findings are largely valid for the overall genre-textbook treatment of literature. We have structured the course so that both groups are together for morning lectures, which emphasize clinical features, molecular pathogenesis, and known potential therapies. According to the 2017 river flood protection assessment, all 3500 km of primary flood defenses must meet a new safety standard by 2050. Currently, 1500 km needs reinforcement. This evaluation aims to meet the new safety rules. Dike failure probability is seen as a mix of mechanisms, with dike slope instability being a key one \u2013 soil movement due to increased groundwater pressure.\n\nHigh water levels lead to heightened groundwater pressure through direct infiltration and subsurface flow. The current dike safety assessment uses an analytical approach for groundwater conditions, neglecting parameters like hydrological attributes, subsurface variability, and flood hydrograph shape. A groundwater model linked with a geomechanical model assessed dike slope stability, varying parameters that influence it via groundwater conditions.\n\nThe significance of subsurface and geometry for dike slope stability under steady-state groundwater conditions was explored through a sensitivity analysis involving fifteen parameters related to geometry, drainage, and material properties. Dike slope and subsurface material type were pivotal factors, with complex connections influencing stability directly through geomechanical traits and indirectly through groundwater conditions. However, this assumes pressure conditions stabilize during high water events, which isn't always accurate.\n\nThe significance of subsurface and geometry for dike slope stability was studied using a sensitivity analysis. Among fifteen parameters related to geometry, drainage, and materials, the dike slope and subsurface material had the greatest impact. Complex connections influence stability through geomechanical properties and groundwater conditions. Yet, this relies on pressure reaching equilibrium during high water events, which may not be consistent.\n\nAnother uncertainty source is the heterogeneity in human-made river dikes and the natural subsurface. DETRIS, an object-based and process-based model, replicates material patterns in river dikes with historical cores. Incorporating heterogeneous DETRIS-simulated dikes into simulations yields a probabilistic assessment that accounts for internal dike heterogeneity. This approach reduces uncertainty by factoring in permeable layers or weak zones.\n\nIn lowland deltas like the Rhine-Meuse delta, the natural subsurface's complexity includes sandy channel belts and clayey floodplain deposits. Current groundwater estimation practices oversimplify to two dimensions, but 2D models can underestimate when the river channel connects in three dimensions. A 2D groundwater model may underestimate dike slope stability compared to a 3D model.\n\nThese uncertain factors merge to influence dike stability. The dike's interior composition significantly affects phreatic levels and heterogeneity. Material composition of the cover layer determines subsurface pressure. This research was conducted with the intention of determining whether or whether not these initiatives considerably enhance the quality of life for current students, faculty members, and alumni, in addition to the communities that are located in the surrounding areas. The data obtained from these university's websites and other sources were supplemented by firsthand accounts obtained by the researcher through the use of a questionnaire and interviews with members of the students, alumni, consultants, faculty of each institution, as well as the two presidents of the universities. This study acknowledged and investigated the positive effects that would result from incorporating South-South international collaboration activities into higher education institutions. It was also shown that IBCs could be able to assist underdeveloped nations in accomplishing some of the Sustainable Development Goals set out by the United Nations. However, iron is a challenging mineral to add to food products. Iron-containing compounds can react with the (phyto)chemicals present in foods and as a result cause severe changes in the organoleptic properties, for instance off-flavor and off-color. To date, iron fortification of foods has proven to be an efficient and cost-effective approach to overcome iron deficiency. Iron-containing compounds that are applied as iron fortificants are divided into three main categories; water-soluble, poorly watersoluble (but soluble in dilute acid), and water-insoluble (and poorly soluble in dilute acid). Although water-soluble compounds have the advantage of high iron bioavailability, the other categories are more in the center of attention because of their minimum influence on the organoleptic properties of the foods which is a consequence of their limited solubilities.\nAmong the poorly water-soluble or water-insoluble iron compounds, ferric pyrophosphate\n(Fe(III)PP) has attracted a great deal of attention. Fe(III)PP is a white\nsolid that prevents addition of unwanted colors to foods. It has been\nshown that Fe(III)PP is very poorly soluble in the food relevant pH (3-7) which is the\nreason for the limited reactivity of this salt with the fortified food vehicle. Furthermore,\nFe(III)PP has low solubility at low pH and enhanced dissolution\nat high pH which is advantageous for ensuring the sufficient\niron bio-accessibility. However, it has previously been shown that addition of iron\nin the form of Fe(III)PP cannot fully prevent the discoloration of the phenolic-rich\nfoods. Therefore, improving the function of Fe(III)PP as an iron fortificant (i.e., decreasing\nthe iron-mediated reactivity while ensuring the iron bio-accessibility) still\nremains of interest. In this work, we seek the strategies by which we can design iron-containing compounds with minimum solubility in the food-relevant pH (3-7), and high and\/or fast dissolution in gastric and intestinal pH (1-3 and 6-8, respectively). Interestingly, mother nature can help us find the answer. Inspired by naturally-occurring minerals such as anapaite (i.e., a mixed calcium\u2013iron phosphate mineral), we intend to embed iron in the matrix of a second (divalent) metal (or mineral) salt, which is less chemically reactive, aiming for: (i) decreasing the iron-mediated reactivity to preserve the organoleptic properties of the food vehicle, and (ii) increasing iron dissolution from the designed multi-mineral salt in the gastric conditions to ensure bio-accessibility of iron (and the other mineral). Once the identification of the prognostic tumour characteristics has been optimized an appropriate therapy can be advised, whereas others may safely choose to refrain from adjuvant therapy. In this thesis we tried to optimize the process of identification of patients with poor prognosis, the efficacy, the safety and the cost-effectiveness of adjuvant therapies need to be addressed separately, and was beyond the scope of this thesis.\n\nIn Chapter 2 we described the prognostic value of microvessel density (MVD) as a surrogate marker for angiogenesis, one of the hallmarks of cancer. As angiogenesis itself remains difficult to measure directly, MVD may provide insight in this microenvironmental process. This computerized morphological study showed that MVD increases with stage, which may explain the observation that stage II patients with high MVD had poor prognosis, biologically being a kind of pre-stage III. Actual stage III patients with high MVD, that were all treated with adjuvant 5-FU based chemotherapy, showed better prognosis on the contrary. We hypothesized that residual or recurrent tumour tissue in stage III patients, from primary tumours with high MVD, were better penetrable for chemotherapy, explaining the improved survival.\n\nIn Chapter 3 we tried to validate CDX2 immunohistochemically, being a promising biomarker published in a high-impact journal by Dalerba et al. Our study showed the practical hurdles involved in validating biomarkers in other cohorts. For disease free survival we were not able to find significant differences between high and low expression of CDX2. However, for disease specific survival we did find comparable results. For a subset of 41 patients we analysed expression of CDX2 by both immunohistochemistry and mass spectrometry. Interestingly, discriminatory power of CDX2 as a prognostic biomarker detected by mass spectrometry outperformed the immunohistochemical detection method. This observation shows that in some cases the biomarker itself may be promising, but the method ought to be fine-tuned to benefit from that marker\u2019s full potential.\n\nIn chapter 4 we presented a new biomarker, obtained from promising research in mice. Expression of KCNQ1 and CD44, both regulated by Wnt-signalling, was analysed immunohistochemically in 386 stage II and III patients, and on mRNA in an external cohort of 90 patients. We concluded that KCNQ1 was a strong prognostic biomarker for disease recurrence. KCNQ1 may be particularly useful in stage II MSS patients, where the question remains which patients are at risk for recurrence and might benefit from adjuvant chemotherapy.\n\nIn Chapter 5 MACROD2 was discussed, a relatively unknown gene, although a tumour suppressing function by activating PARP1 and DNA repair is suspected. We indeed found that low nuclear expression was associated with poor prognosis in stage III MSS colon cancer patients, treated with 5-FU based ACT. Even more, high expression of MACROD2 may serve as a predictive biomarker in stage III MSS tumours, favouring adjuvant treatment with 5-FU compared to no adjuvant treatment at all.\n\nIn the final chapter, Chapter 6, we describe an ultimate attempt to improve the identification of high risk patients. Several biomarkers have been studied on our cohort over the years, some with promising and some with disappointing results. For this study we intended to analyse all these markers combined, to find out which (combination of) biomarkers was able to estimate prognosis best, and whether or not previous less important biomarkers became more interesting. Although other techniques were also used on (some subsets of) this cohort, i.e. mass spectrometry, next generation sequencing and morphological analysis (MVD), for this study we chose for a feasible and widely applicable technique like immunohistochemistry. Therefore, all immunohistochemical biomarkers examined previously on tissue micro arrays were included, in addition to all common clinical and pathological parameters available in our cohort. people's social, political and economic life. Media communication specialists play an important role in shaping public opinion, promoting various products and services, and ensuring interaction between different social groups. In this regard, the importance of high-quality training of professionals capable of working in this field is increasing. The work on training media communication specialists requires an integrated approach that must take into account both internal and external factors. Internal factors are related to the quality of the educational process within universities, the qualifications of teachers at the university, the current state of the material and technical base, as well as the level of motivation of students. External factors include social, economic, technological, and political conditions that directly affect the educational process. The purpose of this article is to analyze the influence of the above factors on the process of training future media communicators in modern universities of the humanities. However, there is no substantive evidence to date that the presence or load of individual species is driving this process. Th is may reflect the complex underlying microbial diversity found in the CF lung at both the inter and intra species level. Therefore, directly sampling of this diversity using high-throughput sequencing will enable major advances in our understanding of APEs. This project involves two approaches which may lead to the discovery of the microbial factors which drive the progression of APE's:(a)Targeted enrichment technology to enable genomic sequencing of multiple CF pathogens concurrently. This w ill enable a greater understanding of the evolutionary dynamics of CF pathogens in the context of APEs. (b)Temporal clonal analysis of Pseudomonas aeruginosa isolates which have been phenotyped for characteristics related to bacterial virulence. Next, I will examine the British Biotech controversy of the 1990s. After exaggerating their production of marimastat for cancer treatment, the company lost accreditation and eventually dissolved. I will explore this public controversy using as a key source the episode in BBC2\u2019s series Blood on the Carpet that recorded the politics behind the company\u2019s downfall as a key source. Finally, I will examine the use of Foucauldian ideas in the rise of British bioethics in the 1960s. Focusing specifically on the emergence of what, following Ludwik Fleck, I call the bioethical thought collective, I hope to expand on Daniel Wilson\u2019s work by analyzing how the collective\u2019s philosophical ideals deployed and engaged with Foucault\u2019s concept of biopower. A critical step in the production of proteins is the translation of nucleotide sequence in messenger RNA molecules into amino acid sequences in proteins. It is well known that protein molecules in turn bind RNA and regulate translation. Other molecules called small regulatory RNAs are often involved in regulating this process: these bind to the messenger RNAs to modulate protein production. Bacteria and other microorganisms use small regulatory RNAs to respond to stress conditions and environmental changes. The ability to rapidly adapt to these changes enables bacteria to withstand harmful conditions, and to use efficiently available resources available; it also helps orchestrate complex programmes of gene expression involved in infection of hosts. For many small RNAs, interactions with messenger RNAs are facilitated by protein chaperones. In some cases the chaperone protects the small RNAs or helps to interact with target RNAs, and then recruits enzymes that eventually degrade the complex formed by the RNA molecules. The project will study the key players involved in the processing and turnover of RNA and the use of RNA to control gene expression in model bacteria. I discovered the fundamental importance of this protein by demonstrating that Eros-deficient mice die from Salmonella infection because their phagocytes cannot make reactive oxygen species. This is because Eros is essential for expression of vital components of the phagocyte NADPH oxidase. My work represents the only paper on this protein. I have found that Eros-deficiency has effects that go far beyond the generation of reactive oxygen species. In particular: \u2022 Eros regulates the expression of other key macrophage proteins including P2X7, a key activator of the NLRP3 inflammasome \u2022 Eros regulates the expression of numerous cytokines from CD4+ T cells. Eros -\/- T cells make 10-fold more IL-4 than control cells In mouse and human systems, I will investigate the molecular mechanisms by which Eros: \u2022 controls the abundance of a subset of proteins working on the hypothesis that it is a novel component of the protein quality control pathway using structural, biochemical and cell biological techniques. \u2022 controls T cell cytokine secretion. However, progress in achieving such behaviour change is slow. Traditional approaches to behaviour change involve providing information with, at best, modest population-level effects and sometimes increased inequalities. Conversely, Choice Architecture interventions (\u201cNudges\u201d) have potentially larger, more equitable effects, involving re-designing environments e.g. reducing plate size to reduce food consumption. However, evidence of effectiveness in real-world settings and understanding of mechanisms are limited. We will bridge this knowledge gap through a novel collaboration between behavioural and cognitive sciences. In the most ambitious co-ordinated set of studies to date, we propose field studies to estimate effect sizes of promising Choice Architecture interventions to reduce food, alcohol and tobacco consumption. Enabled by unprecedented collaborations, these will be conducted in supermarkets, bars and cafeterias and interventions optimised through laboratory studies determining mechanisms. We will run international workshops, public engagement activities and a Behaviour Change Summit to facilitate implementing the evidence generated, overseen by an Implementation Advisory Panel. Because these unhealthy behaviours are more common amongst the poorest, they contribute to the gaps in health and life expectancy between the richest and the poorest. Crystallographic methods in our new computer program Phaser are based on the principle of maximum likelihood, which requires an understanding of the statistical relationships among experimental observations. We will build on success in the area of molecular replacement to solve structures with less prior knowledge, usin g either smaller fragments or ab initio models. We will complete the implementation of experimental phasing by multiple-wavelength isomorphous replacement and two-wavelength anomalous diffraction, and we will explore other applications, for instance to phasing in the presence of radiation damage. In our structural research, we will complete our study of the enzymatic mechanism of pertussis toxin by determining structures representing the Michaelis complex. We will study the mechanism by whi ch the hormone-binding globulins carry out a targeted release of bound hormone, and will attempt to exploit that understanding to design variants that release selected drugs at selected target sites. FACS isolation of LGR5-positive intestinal stem cells have facilitated the expression profiling of LGR5+ stem cells, which established stem cell-specific genes of the adult intestine. Amongst are two homologous E3 ubiquitin ligases, RNF43 and ZNRF3, the inactivation of which resulted in unrestricted expansion of intestinal stem cells and formation of adenomas due to hyper-activat ion of Wnt signalling. This has started uncovering the importance of E3 ligases in regulating intestinal stem cells. Besides RNF43 and ZNRF3, additional uncharacterized E3 ubiquitin ligases are enriched in intestinal stem cells. By using cell-surface marker CD24 to sort and profile Paneth cells, novel additional E3 ubiquitin ligases were also detected in these niche cells. These novel E3 ubiquitin ligases are the focus of this research plan, since it is anticipated that they play an important ro le in stem cells, niche cells and their interaction.
